Epigenetic regulation of the function of genes and genome is an important mechanism in the regulation of vital functions of cells and the body and is complicated by inflammation in parodontal tissues against the background of metabolic syndrome. Purpose of the study. Study of methylation of IFNγ gene promoter in chronic generalized parodontitis of different degree on the background of metabolic
syndrome. Materials and methods. In 21 patients, DNA was amplified in the blood and in the tissues of the gums. Pyrosequencing was carried out using PyroMark Gold Q24 Reagents on the PyroMark Q24, according to the Qiagen protocol. The content of methylated DNA in the sample was assessed using PyroMark CpG software 2.01. Results. Conclusions. The decrease in the parodontal tissues of the
methylation of the IFNγ gene promoter on the background of the metabolic syndrome can be a marker of the activity of the chronic inflammatory process in the parodontium. The observed increase in IFNγ gene expression in periodontal tissues on the background of the metabolic syndrome ultimately contributes to the progression of parodontitis and destruction of bone tissue.
Показано, что снижение в тканях пародонта метилирования промотора гена IFNγ на фоне метаболического синдрома у пациентов с хроническим генерализованным пародонтитом II-III степени, по сравнению с показателями у пациентов с хроническим генерализованным пародонтитом начальной – I степени, может быть маркером активности хронического воспалительного процесса в пародонте. Наблюдаемое повышение экспрессии гена IFNγ в тканях пародонта при метаболическом синдроме, в конечном итоге, способствует прогрессированию пародонтита и разрушению костной ткани.
