<title>Different mechanisms of axitinib and diazepam antiseizure action in pentylenetetrazol-induced kindling model</title>

<description>Different mechanisms of axitinib and diazepam antiseizure action in pentylenetetrazol-induced kindling model
Godlevsky, L. S.; Is¸ınsu Alkan; Kıymet Kübra Tüfekci; Pervak, M. P.
Introduction: This study intended to assess the histomorphology characteristics of hippocampal structures and&#13;
determine the severity of seizures after treatment with the tyrosine kinase B inhibitor axitinib and diazepam in&#13;
fully developed and postponed period in PTZ-kindled rats.&#13;
Methods and materials: Wistar rats were given PTZ for 21 days until fully developed convulsions were achieved.&#13;
Two protocols were explored: assessment of seizures immediately after the completion of the kindling (early&#13;
kindling) and after a two-week post-kindling PTZ-free period. Treatment with axitinib and diazepam was performed before early and postponed kindling seizures assessment, with the consequent collection of brains for&#13;
histomorphology.&#13;
Results: Axitinib and diazepam effectively reduced seizure severity at the early and postponed periods of kindling.&#13;
Axitinib’s antiseizure effectiveness was reduced in the postponed stage of kindling period compared with the&#13;
early one (P = 0.039), while diazepam’s effectiveness was maintained at a similar level (P &gt; 0.05). Stereological&#13;
quantification of neuronal hippocampus changes revealed an increase in the total volume of the stratum radiatum, while a decrease of the dentate gyrus, in postponed compared with early kindling (P &lt; 0.05). The positivity of collagen type IV, which is present in the blood-brain barrier, increased and was more pronounced in the&#13;
postponed period in hippocampal structures (CA1-CA3).&#13;
Conclusion: The antiseizure effect of tyrosine kinase B inhibition with a specific antagonist of VEGF axitinib is&#13;
particularly pronounced in the early phase of PTZ kindling. Opposite to axitinib, diazepam demonstrated a&#13;
similar antiseizure action in both periods of kindling. These results suggest a more pronounced contribution of&#13;
angiogenesis compared with the neuronal degenerative hippocampal damage to the development of PTZ-kindled&#13;
chronic epileptogenesis.

<description>On different mechanisms of axitinib and diazepam antiseizure action in pentylenetetrazol-induced kindling model
Godlevsky, L. S.; Süleyman, K.; Pervak, M. P.
PTZ-induced kindling exhibits resistance to antiepileptic medication, increased brain-blood barrier. Tyrosine kinase inhibitors demonstrated antiepileptic efficacy, suggesting that VEGF-induced angiogenesis plays a part in the&#13;
development of persistent epileptic activity. This study used PTZ-kindled rats to assess the histomorphology features of the hippocampus structures and ascertain the degree of seizures following treatment with axitinib and diazepam, tyrosine kinase B inhibitors, throughout fully developed and postponed phases.

<title>Dataset on quantitative behavioral and EEG of lateral cerebellar nucleus (LCN) electrical stimulation in PTZ-kindled rats</title>

<description>Dataset on quantitative behavioral and EEG of lateral cerebellar nucleus (LCN) electrical stimulation in PTZ-kindled rats
Godlevsky, L. S.; Pervak, M. P.; Yehorenko, O. S.; Marchenko, S. V.
The data represented here are in relation to the manuscript&#13;
(Godlevsky et al., 2025) [1], which quantitatively estimated&#13;
the effects of the lateral cerebellar nucleus (LCN) electrical&#13;
stimulation (100 Hz) in rats kindled with pentylenetetrazol&#13;
(PTZ) daily administration. Both behavioral Racine scale scoring seizure severity and EEG seizure discharges registered in&#13;
the frontal cortex of rats kindled to the early stage of kindling (not exceeded 2 scored seizure severity) after 9-11 PTZ,&#13;
and fully developed kindling (4-5 scored seizure severity) after 21 PTZ were quantitatively analyzed. The data in the Tables favors the contribution of LCN to the dynamic of behavioral and EEG seizures, namely, the facilitation of myoclonic&#13;
seizures at an early stage and the prevention of generalized&#13;
seizures at the stage of kindled tonic-clonic seizures. Accordingly, all results are presented in primary individual form and&#13;
quantitatively analyzed, which might better understand LCN’s&#13;
role in the pathogenesis of chronic epileptic syndrome.

Arabadzhy, D. R.; Ryabenka, O. D.; Shcheglov, I. A.; Prybolovets, K. O.; Kashchenko, O. A.; Bidnyuk, V. K.
Our work aimed to produce kindled seizures with transcorneal 6‐Hz electrical stimulation (ES) (32 mA 6 Hz for 3 seconds (GRASS stimulator, Astro.Med.Inc., USA) that was performed daily 5 times per week. Al‐ together 25 stimulations were delivered to each rat. During ES, clonic seizures of body muscles were seen with trouble tails that were stopped immediately after ES cessation.

<description>Rapamycin combined with pitolisant alleviates anxiety and depression in pentylenetetrazol (PTZ) kindled rats
Pervak, M. P.; Poshyvak, O. B.; Yehorenko, O. S.; Arabadzhy, D. R.; Marchenko, S. V.
The work aimed to investigate the anxiety and depression manifestations in PTZ‐kindled rats under combined treatment with the rapamycin and histamine H3 receptor inverse agonist pitolisant. Kindling was produced in Wistar male rats by administration of three‐week PTZ (35.0 mg/kg, i.p.). Treatment with rapamycin (Pfizer, 0.5 mg/kg) and pitolisant (Selleck, 5.0 mg/kg) was performed for ten days in fully kindled rats. Control rats were treated with DMSO. Kindled an‐ imals spent less time (2.7 times) in the open area of the elevated plus maze (EPM) in comparison to the control (P&lt;0.001). F

<description>Antiseizuree effectiveness of nicotinamid and diazepam increased after brain transcranial direct current stimulation (tDCS)
Prybolovets, K. O.; Pastuhov, O. O.; Budigay, N. S.; Kibysh, Ya. A.; Godlevsky, L. S.
tDCS of the cerebellum prevented PTZ‐kindled seizures, and such an effect was strengthened after diazepam administration. The work aimed to investigate the pronouncement of antiseizure effects of nicotinamide and diazepam performed after preliminarily applying tDCS to the cerebellar or forebrain of PTZ‐kindled rats.

Manifestations of metabolic syndrome (MS) man‑&#13;
ifestations in chronic epilepsy models were in the&#13;
scope of the investigation. Kindling was induced by&#13;
daily injections of PTZ at a dose of 35 mg/kg i.p. for&#13;
three weeks. Those animals with fully developed gen‑&#13;
eralized seizures were used for investigations. Glucose&#13;
and insulin levels were not different from control rats,&#13;
while PTZ‑kindled rats demonstrated decreased tol‑&#13;
erance to glucose tests.
